Im pretty sure that's not possible. There is no option on the email entity itself for this type of a request, unless you could set all emails from the exchange server (if you have one), it wouldn't be a CRM specific setting. You might try the Microsoft Exchange forum to see if you can force a read-receipt for all emails. Additionally, if you did somehow send the email with a read-receipt, without a forward mailbox or inbound rule you'd never get the response back into the CRM server. Hope that helps.
