Hi,
Wondering if anyone can tell me if Email Router and Rule Deployment is
required for the automatic generation of campaign response to work in CRM4?

The e-mail router and rule deployment is one of the methods for e-mai
integration in CRM as described in the implementation guide

...but it is not necessary to have the e-mail router installed to have
campaign responses automatically created and tracked in CRM as described here
in this blog

You DO however have to set the "Track e-mail option" to Yes on the E-mail
tracking tab on the system settings dialog box, and verify that campaign
responses for incoming E-mail is set to Yes on the Marketing tab in that
dialog box.

Your answer is what I beleived at first. I have all those options setup as
per your understanding below, but no Campaign Response is being generated, I
tested from 3 seperate environment and they all reacts the same way, no auto
Campaign Response. Is anyone is using this function in CRM4 and can you tell
me if it is working? Do you have Event Accelerator installed? I am thinking
if it is the Event Accelerator broke this function? Or maybe Rollup 6 broke
this function?
